I was working on my UNIX workstation when I had the bright idea to remove the CPU cooler for dusting purposes. Do I now need to replace the thermal paste before I use it again?

If there's still lots there and it's not all dried up, don't worry about it.

But here's the deal: The paste is old and needs to be replaced anyway. Can I use it until Wednesday, when I will have time to go out and buy some thermal paste, even though I removed the cooler?

As long as temps are low no problem.

Shut that thing down and DO NOT use it until you reapply a fresh coat of thermal paste. Anything over 90 degrees C has the potential to damage poorly connected or impure solder areas, and will cause massive thermal wear and tear on the CPU even if it does keep running.
Also remember to clean off all the old thermal paste prior to applying a fresh coat, and make sure that it's applied per manufacturer instructions.
